VectorBuilder has announced a platform collaboration with Ireland-based Lir Therapeutics to engineer AAV gene therapy capsids optimised for both biological performance and manufacturability, giving gene therapy developers a clearer route to capsids designed for GMP-scale production from the outset rather than re-engineered for manufacturing after candidates are already selected.
The collaboration combines VectorBuilder's DeepCap capsid discovery platform and GMP manufacturing capabilities with Lir's AI-driven nAAVigator computational protein engineering platform, with Lir applying nAAVigator to lead capsids identified through DeepCap rather than starting development from entirely new vectors.
The companies will initially evaluate several VectorBuilder capsids across multiple therapeutic applications, with VectorBuilder responsible for process development and GMP manufacturing and Lir providing computational engineering and experimental validation. No specific indications have been disclosed, and financial terms were not disclosed.
VectorBuilder, headquartered in Chicago and founded in 2014, is a global gene delivery and CDMO company offering vector design, cloning and GMP manufacturing of clinical-grade plasmids, mRNAs, proteins and viruses; it is separately investing $50m to expand its Chicago biomanufacturing and R&D centre.
Lir Therapeutics, founded in 2024, is an AI viral vector company developing nAAVigator, an AI-driven capsid design platform aimed at addressing manufacturing, dose, tissue-targeting and immune-evasion limitations in AAV-based gene therapies; the company has previously received investment from the Cell and Gene Therapy Catapult's Innovate UK Cross Catapult Investment Pilot.
The collaboration reflects a structural shift in AAV capsid engineering towards co-optimising for manufacturability alongside biological performance, as production yield and process reproducibility have become the binding constraint on moving gene therapies from research into clinical and commercial scale.
AI-guided capsid discovery has become a competitive field, with Dyno Therapeutics running parallel collaborations with Roche and Astellas, while VectorBuilder's own DeepCap platform already underpins a separate option agreement with Ikarovec potentially worth more than USD 1 billion.
For the sector, the deal reinforces that GMP manufacturing capability, not discovery science alone, is becoming a distinct value driver in gene therapy licensing and platform partnerships.
The wider signal for the sector is that platform partnerships pairing AI-driven capsid design with CDMO-scale GMP manufacturing capacity are becoming the preferred structure for de-risking gene therapy production, rather than a downstream afterthought.
